Abstract

One of the genes that can regulate the traits and quality of milk production in cattle is the Pituitary-Specific Transcription Factor-1 (Pit-1) gene. The detection of variations in the Pit-1 gene at the Pit-1-Hinf locus is expected to provide insights into the potential of Aceh cattle to become dairy cattle and serve as a guide in selecting dual genetic traits in Aceh cattle. The samples used were 8 blood samples consisting of 4 male Aceh cattle and 4 female Aceh cattle as sources of DNA taken from local Aceh cattle in several districts. The method used was (PCR-RFLP), starting with DNA extraction, PCR, and then amplification of DNA with the restriction enzyme HinfI. to ascertain allele variants and the PIT1 genotype. The genotype and allele frequencies are computed using the genotyping procedure. The study’s findings indicate that there is just one type of PIT1 genotype, BB, with a frequency of 1. The alleles found are B and B, each with a frequency of 1. This study’s findings lead to the conclusion that there is no genetic diversity of Pit-1|Hinfl in Aceh cattle, as only the BB genotype was found. This indicates that selected Aceh cattle remain in pure condition as beef cattle types and have not shown their potential as dairy cattle types. Thus, Pit-1|Hinfl cannot yet be used as a genetic marker to search for Aceh cattle as dairy cows with a small sample size; however, Pit-1|Hinfl can be used on Aceh cattle if a larger sample size is used, allowing for diversity and obtaining further information.
