Abstract
Pleurotus sajor-caju strains (PscW and PscA), indigenous to Wli forest in the Volta region and Atewa forest in the Eastern region of Ghana, respectively, were studied to identify optimal cultivation conditions. This research aimed to determine the ideal temperature, pH and growth media for the vegetative development of these oyster mushrooms strains. Mycelia growth was evaluated across varying temperatures (26°C, 28°C and 30°C), pH levels (6, 6.5 and 7) and media types, including malt extract agar (MEA) and potato dextrose agar (PDA). Additionally, substrates such as sawdust, rice straw, maize straw and cassava peels were incorporated into the media to assess their impact on mycelial morphology. The study found that MEA at 30°C provided the most favourable conditions for mycelia growth, recording radial mycelial growths of 3.80 ± 0.04 cm and 3.88 ± 0.09 cm for strain PscW and PscA, respectively, on Day 5. Furthermore, rice and maize straw were identified as effective components in the preparation of starter cultures for these mushroom strains. The successful cultivation of these mushrooms on agricultural wastes demonstrates their potential as alternative nutrient sources for vegetative growth and supports their suitability for sustainable mushroom production in Ghana.
