Effect of ulinastatin on the hyperpermeability of pulmonary vessel induced by lipopolysaccharide and the expression of vascular endothelial cadherin

Abstract

Objective:To observe the effect of ulinastatin(UTI)on the hyperpermeability of pulmonary vessel induced by lipopolysaccharide(LPS)and vascular endothelial cadherin(VE-cadherin)expression.Method:Forty C57BL/6mice were randomly divided into normal control group(CON group);ALI model group(LPS group);UTI single use group(UTI group);UTI treatment group(UTI+LPS group).Each group consisted of 10 mice.In CON group,saline 0.2ml was intraperitoneally injected respectively;in ALI group,LSP 10mg/kg was intraperitoneally injected respectively;in UTI group,UTI 50 000 U/kg was intraperitoneally injected respectively;in UTI treatment group,LPS 10mg/kg+UTI 50 000U/kg was intraperitoneally injected respectively.After 12 hours,the permeability of pulmonary vessel were observed by Evans Blue method.VE-cadherin expression in lung were detected by Western blot respectively.Result:After 12 hours,LPS increased the level of the eluted Evans Blue in lung tissue(P0.01)and decreased VE-cadherin expression in LPS group compared with those of the CON group(P0.01).UTI attenuated the level of the eluted Evans Blue in lung tissue(P0.01)and increased VE-cadherin expression(P0.01)in UTI+LPS group compared with those of the LPS group.Conclusion:UTI may upregulate VE-cadherin so as to maintain the normal permeability of pulmonary induced by LPS.
