Influence of icotinib hydrochloride on apoptosis of human tongue carcinoma cell line Tca8113 and expressions of Bcl-2 protein and Bax protein

Abstract

Objective To study the effect of icotinib hydrochloride on the apoptosis of human carcinoma of tongue Tca8113 cell line and the expressions of Bcl-2 and Bax protein in order to explore the mechanisms.Methods Tca8113 cells were incubated with icotinib hydrochloride at a dose of 0,10,20,40 μmol·L-1 for 24,48,72 h respectively.Apoptosis were detected by flow cytometer(FCM).Immunocytochemical method,indirectly immunofluorescence and FCM were used to detect the expression of the Bcl-2 and Bax protein after Tca8113 cells were treated with icotinib hydrochloride.Results After the treatment of icotinib hydrochloride,the Bcl-2 protein expression decreased and the Bax protein expression increased of human carcinoma of tongue Tca8113 cells.The percentage of apoptotic cells was gradually increased with the increasing of concentration and treating time of icotinib hydrochloride.The percentage of apoptotic cells were positive correlated to the concentration and treated time of icotinib hydrochloride.Conclusion Icotinib hydrochloride can decrease the expression of the Bcl-2 protein and increase the expression of the Bax protein,and it can induce apoptosis of human carcinoma of tongue Tca8113 cell.
