tlooto Summary
Water-soluble polysaccharide isolated from Atractylodes macrocephala Koidz exhibits a molecular weight of 8374 and a basic structure composed of Glc, Rha, and Man.
Abstract
The water soluble crude polysaccharide was seperated from Atractylodes macrocephala Koidz by water. After ethanol precipitation,Sephadex G-75 gel filitration, the fraction of AMP was obtained.By analyzing of Gel filtration on Sephadex G-75,ion-exchange chromatography on DEAE column and HPLC,the homogeneous of AMP was determined. The molecular weight of the AMP is 8 374 measured by HPLC.GC analysis indicated that AMP was composed of Glc,Gal,Rha and Man,the molar ratio was 7.36∶1.00∶3.05∶1.52.By analyzing the products of partial hydrlsys,Smith degradation, methylation and GC-MS analysis, the basic structure of AMP was determined as Glc,Rha and Man.On average, there was 1 branch among 6 main chain residues, which was substituted at 4-O of Glc and 6-O of Man.The nonreduced end was Glc.
