Differential Proteomic Studies on Pollen Grain Proteins of Wheat Male Sterile Line Induced by Chemical Hybridizing Agent SQ-1

Abstract

The pollen grain proteins in maturation-stage from wheat(Triticum aestivum) male sterile line induced by chemical hybridizing agent SQ-1 were separated by 2-D electrophoresis with immobilized pH(4~7) gradients as first dimension and SDS-PAGE as second dimension.The silver-stained protein spots were analyzed using PDQuest software,over 350 protein spots were detected reproducibly within pI 4~7 on each 2-D gel.Twenty-one spots were found to be differentially expressed.Seven differentially expressed protein spots out of 11 were identified by MALDI-TOF mass spectrometry analysis and protein database searching,which are vacuolar invertase,ascorbate peroxidase,putative fructose-1,6-biphosphate aldolase,dynein light chain TCTEX-1,lectin protein kinase,manganese superoxide dismutase and predicted protein.The functions of major differentially expressed proteins were reviewed and discussed.It was inferred that the male sterility induced by SQ-1 might related to energy metabolism turbulence,starch synthesis suffocation,active oxygen accumulation,apoptosis and disturbed function of the flower developmental gene.
