A case-control study of polymorphism of XRCC1 gene and the risk of hepatocellular carcinoma

Abstract

Objective To approach the distribution frequency and characteristics of 3 common SNP loci of X-ray repair cross complementing group 1 (XRCC1) in subjects and investigate the relationship between the codon 194TT,280AA,399AA of XRCC1 and Hepatocellular Carcinoma (HCC). Methods The research was designed as a hospital-based case-control study. 3 common SNP loci of XRCC1 gene were measured with TaqMan MGB,a fluorescent quantitative real time PCR method. Results (1)The genotype analysis showed that allele mutation frequency of gene sites of XRCC1-194,XRCC1-280,XRCC1-399 in the control group were 27.1%,9.2%,23.6% respectively,with no statistically significant difference with the case group (25.7%,9.3% and 24.8%) (all P0.05). Also their genotype distribution showed no statistic difference. (2)Multiple factors non-conditional Logistic regression analysis indicated that after adjusted for age,sex,nationality,marriage,occupation,there was no evidence of the relationship between XRCC1-194TT,XRCC1-280AA and XRCC1-399AA of XRCC1 and HCC. (3)Interaction analysis of gene polymorphism and environment factors indicated there existed interactions between these gene polymorphism and environment risk factors,such as smoking,drinking,chronic HBsAg infection. Conclusions XRCC1-194TT,XRCC1-280AA and XRCC1-399AA of XRCC1 may be not associated with the risk of HCC in GuangXi. There may exist interaction between gene polymorphisms of XRCC1 and the HCC risk factors like smoking,drinking and chronic HBsAg infection.
